We have found that a member of the nuclear hormone receptor superfamily, the peroxisome proliferator-activated receptor a (PPAR a ), serves as a key regulator of mitochondrial energy production in the normal postnatal mammalian heart. The activity of PPAR a is reduced in the hypertrophied and failing heart. Conversely, PPAR a activity is abnormally super-activated in the diabetic heart. Chief Scientist Yuichiro Maeda and his colleagues in the Structural Biochemistry Laboratory have become the first in the world to solve the three-dimensional structure of troponin. Troponin is one of the proteins that compose actin filaments, and it plays an important role in the regulation of muscle contraction. This landmark achievement had been awaited for 40 years, since the proposal of the calcium ion (Ca ) theory of regulation of muscle contraction and the discovery of troponin, a molecule to which Ca binds. "Our goal is to first elucidate the three-dimensional structure of actin filaments, and then to use the knowledge to fully clarify the regulatory mechanisms of muscle contraction. Now that we have determined troponin's three-dimensional structure, it is possible to get to the heart of the questions concerning troponin's behavior and functions," explained Dr. Maeda. Extractions: A heart attack (also known as a myocardial infarction) is the death of heart muscle from the sudden blockage of a coronary artery by a blood clot. Coronary arteries are blood vessels that supply the heart muscle with blood and oxygen. Blockage of a coronary artery deprives the heart muscle of blood and oxygen, causing injury to the heart muscle. Injury to the heart muscle causes chest pain and pressure. If blood flow is not restored within 20 to 40 minutes, irreversible death of the heart muscle will begin to occur. Muscle continues to die for 6-8 hours at which time the heart attack usually is "complete." The dead heart muscle is replaced by scar tissue. Approximately one million Americans suffer a heart attack each year. Four hundred thousand of them die as a result of their heart attack.

Hypothesis: The aim of this study was to assess the possible involvement of apoptotic process in the pathogenesis of DCM and myocarditis. Methods: Endomyocardial biopsy was performed in patients with DCM (n = 9), myocarditis (n = 4), or atypical chest pain syndrome (as controls; n = 5). The TUNEL method was used for in situ detection of oligonucleosomal DNA strand breaks. Results: Conclusion: The DNA strand breaks were detected in cardiomyocytes in patients with DCM and mainly in interstitial cells in myocarditis. It is possible that the DNA strand breaks can be involved in mechanisms of progressive loss of functional cardiac units in these myocardial diseases.
