Warning Signs in Tuberous Sclerosis
Introduction
The following are some things that can act as warning signs of serious problems in Tuberous Sclerosis. Many of them can occur for a variety of other reasons that may be more trivial but they can all be a warning of a much more serious problem and should be discussed with your doctor. Further information and advice can also be obtained through the Tuberous Sclerosis Association and their panel of expert advisers.
Development and Behaviour
Pre-School Development
Problems with development and behaviour are very common in Tuberous Sclerosis and can be helped by early pre-school programmes as soon as late milestones are noticed. Delayed speech is a particular problem in TS and should lead not only to speech therapy but also to an assessment for autism which can be done from 18 months of age. Help with autism and with concentration problems should not be left until the child starts school. Further help should be sought if you have any suspicion of delayed speech, behaviour problems or if your child seems to be in a world of their own.

Microsoft Closes Activation Loophole Written by Jules Friday, 25 February 2005 By Nate Mook and David Worthington, BetaNews February 24, 2005, 8:05 PM Microsoft is closing a loophole that enabled unscrupulous resellers to use Windows XP product keys that were stolen from large OEMs. The result: customers who purchase Windows on a new PC will not be able to activate, nor reinstall their operating system without first calling Microsoft. The problem lies in the Certificate of Authenticity (COA) labels on PCs, which often contain unused unique product keys because OEMs preinstall Windows and bypass product activation. These keys could easily be copied and sold by a smaller computer dealer complete with a counterfeit COA. Because the product key was never actually used by an end-user, a customer would have no trouble activating Windows via the Internet and never know the difference.

Tuberous sclerosis (TSC) is a rare genetic disease that causes benign tumors to grow in the brain and on other vital organs such as the kidneys, heart, eyes, lungs, and skin. It commonly affects the central nervous system. In addition to the benign tumors that frequently occur in TSC, other common symptoms include seizures, mental retardation, behavior problems, and skin abnormalities. TSC may be present at birth, but signs of the disorder can be subtle and full symptoms may take some time to develop. Three types of brain tumors are associated with TSC: cortical tubers

Community TSC Overview Science Targets Drug Development Visualization ... Corporate Support
Target 1: PTEN
Target 2: P13K
Target 3: FRAP (=mTOR)
Target 4: EIF4E
Target 5: Akt (=PKB)
Overview
The Rothberg Institute is funding some of the best researchers at Harvard, Yale, and the Fox chase cancer center to find new targets for intervention in the treatment of TSC. Those targets are fed into drug development programs right at the ICCB at Harvard, as well at the Rothberg Institute. Read more about libraries of chemical compounds. (PDF) 05/2002
The science of using computers to screen for potential drugs from large collections of candidates against targets that have had their structures determined is known as "Virtual Library Screening". Read more about the science of Large Scale Virtual Screening. (PDF) 01/22/2001 Since D2OL™ is actually a computing and graphical framework that allows us to use the best software as it is created anywhere in the world to do drug design, we will be both improving the Virtual Screening programs as well as adding programs to do atom-by-atom "de novo" drug design.
In addition to taking existing small molecule compounds found in chemical libraries and fitting them to targets that we discover are related to TSC we plan on taking a parallel approach based on making new drugs atom-by-atom to fit into the targets we discover. While this approach seems like science fiction, recent advances by a new member or our scientific advisory board suggest that this approach may have significant advantages of any approaches taken to date. So stay tuned....
