For allergic, seasonal rhinitis, what is the efficacy of antihistamines versus nasal corticosteroids? Antihistamines versus immunotherapy? Nasal steroids versus immunotherapy? Sedating versus nonsedating antihistamines? Other agents? What are the side effects/adverse events due to drug treatments? How do efficacy and side effects of treatments vary by severity of rhinitis, patient chracteristics? How does the treatment of allergic rhinitis impact on the development of asthma? ABSTRACT Background Encasing bedding in impermeable covers reduces exposure to house-dust mites, but the clinical benefit of this intervention as part of mite-avoidance measures for patients with allergic rhinitis is not known. We performed a multicenter, randomized, placebo-controlled trial of one year of use of impermeable bedding covers in the bedrooms of patients with rhinitis who were sensitized to house-dust mites to determine the effects on the signs and symptoms of disease. Methods Three participating university medical centers enrolled 279 patients with allergic rhinitis who were randomly assigned to receive impermeable or non-impermeable (control) covers for their mattress, pillow, and duvet or blanket. At the start of

Lovelace Respiratory Research Institute (LRRI) offers the Beagle dog model of asthma, allergic rhinitis, and allergic dermatitis for studies of drug efficacy and pharmacology. Dogs treated with ragweed or house dust mite extract develop sensitive airways that reversibly constrict. Dogs maintain allergic response over time after repeated exposures to ragweed. Sensitized dogs also develop inflammatory and other allergic responses in the lung (elevated total and specific IgE; eosinophils). Some common asthma medications such as albuterol and oral steroids effectively relieve the airway constriction caused by a ragweed exposure. Finally, acoustic rhinometry can assess nasal obstruction in the dog model after a challenge of ragweed. Allergy drugs such as d-pseudoephedrine block ragweed-induced nasal congestion. CHARACTERISTICS OF OUR DOG MODEL:
    • Natural development of allergic disease Sensitization of ragweed and dust mites IgE-mediated sensitivity Immediate airway responses to allergen Chronic inflammation Localized immune response in the lung
    EXPERIMENTAL CAPABILITIES:
    • Eosinophils: Effects of anti-inflammatory drugs on recruitment and activation of eosinophils Cytokines: IL-2, -4, -8, -10,-12, -13, INF
