Current Research Interests
The Kelly laboratory is interested in regulatory mechanisms related to the control of cardiac energy metabolism in the developing and diseased heart. We have found that a member of the nuclear hormone receptor superfamily, the peroxisome proliferator-activated receptor a (PPAR a ), serves as a key regulator of mitochondrial energy production in the normal postnatal mammalian heart. The activity of PPAR a is reduced in the hypertrophied and failing heart. Conversely, PPAR a activity is abnormally “super-activated” in the diabetic heart. To model these disease states, genetically modified mice with reduced or increased cardiac PPAR a function have been developed. Using the PPAR

Description: This text provides a look into the advances in the fields of molecular biology, development and genetics in relation to cardiology. It offers to the reader an introduction to molecular cardiology via a section of introductory overviews. The authors provide chapters on disease-oriented topics written by leaders in the field. These experts have gathered a concise and thorough discussion of the relevant molecular, cellular, genetic and developmental biological processes that underlie and contribute to cardiovascular disease. This book presents a reference that any scientist in the field would want to own as a broad overview of the field. Also a valuable tool to any cardiology fellow would benefit from due to the insights it provides into the common diseases in the field.

Zeroing in on the regulatory mechanism of muscle contraction
An approach focusing on the three-dimensional structure and movement of actin filaments RIKEN Harima Institute
Structural Biochemistry Laboratory
Chief Scientist, Dr. Yuichiro Maeda Great progress has been made in the research of "actin filaments," which play a part in the regulation of muscle contraction. Chief Scientist Yuichiro Maeda and his colleagues in the Structural Biochemistry Laboratory have become the first in the world to solve the three-dimensional structure of troponin. Troponin is one of the proteins that compose actin filaments, and it plays an important role in the regulation of muscle contraction. This landmark achievement had been awaited for 40 years, since the proposal of the calcium ion (Ca ) theory of regulation of muscle contraction and the discovery of troponin, a molecule to which Ca binds. "Our goal is to first elucidate the three-dimensional structure of actin filaments, and then to use the knowledge to fully clarify the regulatory mechanisms of muscle contraction. Now that we have determined troponin's three-dimensional structure, it is possible to get to the heart of the questions concerning troponin's behavior and functions," explained Dr. Maeda. How can structural biology uncover the regulatory mechanisms of muscle contractions? This article will take you to the forefront of cutting-edge research.

Heart Attack
(Myocardial Infarction)
Revising Medical Author: Dennis Lee, MD
Revising Medical Editor: Daniel Kulick, MD Jay Marks, MD What is a heart attack?
A heart attack (also known as a myocardial infarction) is the death of heart muscle from the sudden blockage of a coronary artery by a blood clot. Coronary arteries are blood vessels that supply the heart muscle with blood and oxygen. Blockage of a coronary artery deprives the heart muscle of blood and oxygen, causing injury to the heart muscle. Injury to the heart muscle causes chest pain and pressure. If blood flow is not restored within 20 to 40 minutes, irreversible death of the heart muscle will begin to occur. Muscle continues to die for 6-8 hours at which time the heart attack usually is "complete." The dead heart muscle is replaced by scar tissue. Approximately one million Americans suffer a heart attack each year. Four hundred thousand of them die as a result of their heart attack.

Breakthrough In National Diseases: Common Factor Behind Myocardial Infarction, Rheumatism And MS
A common gene variant has been identified as the risk factor behind a number of common diseases by research scientists at Karolinska Institutet and the Centre for Molecular Medicine (CMM), Stockholm, Sweden. Up to a quarter of the population could be affected. Related News Stories (June 30, 2005) Parents whose children are born with a low birth weight run greater risk of dying of cardiovascular diseases. Myocardial DNA Strand Breaks Are Detected in Biopsy Tissues from Patients with Dilated Cardiomyopathy
Yukihiko Seki, M.D., Hisashi Kai, M.D., Ph.D.,* Mamiko Kai, Ph.D.,* Akihiko Muraishi, M.D., Kyo Adachi, M.D., Ph.D., Tsutomu Imaizumi, M.D., Ph.D. Third Department of Internal Medicine and *Institute of Cardiovascular Disease, Kurume University School of Medicine, Kurume, Japan Summary Background: Progressive damage of cardiomyocytes with interstitial and replacement fibrosis accompanied by less inflammatory cell infiltration is observed in patients with dilated cardiomyopathy (DCM), suggesting some other mechanisms rather than necrotic cell death. Hypothesis: The aim of this study was to assess the possible involvement of apoptotic process in the pathogenesis of DCM and myocarditis. Methods: Endomyocardial biopsy was performed in patients with DCM (n = 9), myocarditis (n = 4), or atypical chest pain syndrome (as controls; n = 5). The TUNEL method was used for in situ detection of oligonucleosomal DNA strand breaks. Results: Conclusion: The DNA strand breaks were detected in cardiomyocytes in patients with DCM and mainly in interstitial cells in myocarditis. It is possible that the DNA strand breaks can be involved in mechanisms of progressive loss of functional cardiac units in these myocardial diseases.
