What Are Mycoplasmas?
Mycoplasmas are wall-less bacteria belonging to various genera within the class Mollicutes. These flexible, pleomorphic organisms can be as small as 0.2-0.3um and can achieve very high densities in cell cultures, (10 organisms/mL), without discernable pH changes or turbidity. Mycoplasmas can usually be grown on artificial media, but most species require very complex, nutritionally enriched media and carefully defined environmental conditions. There are currently more than 183 species in 8 genera, many of which are pathogenic. The vast majority of cell culture contaminants belong to only 6 species of human, bovine or porcine origin. These very common contaminants are: Mycoplasma hyorhinis, Mycoplasma arginini, Mycoplasma salivarium, Mycoplasma orale, Mycoplasma fermentans and Acholephasma laidlawii.

The two biggest problems in cell culturing are misidentification or cross-contamination of cell lines and contamination with microorganisms (e.g. bacteria, fungi, yeast, viruses and above all mycoplasmas). In order to exclude misidentified and contaminated cell lines and to ensure the authenticity of the cultures, we make efforts to perform a number of analyses on cell lines:
  • viability and growth tests for the detection of microorganisms (mycoplasmas, other bacteria, fungi, yeast) tests for the detection of viral contamination PCR analysis for the determination of murine species DNA fingerprinting karyotyping and cytogenetic analysis immunofluorescence staining of cellular antigens.

  • In addition to this standard quality and identity control program, cell lines are characterized in detail with regard to various parameters at the DSMZ; for details see Quality Control, Identity Control, and Characterization of Cell Lines . Those tests that were performed on any given cell line are indicated for each cell line in the individual entries. The specific tests to exclude cross-contamination and mycoplasma contamination are described in detail under Cytogenetics DNA Fingerprinting , and Mycoplasma Detection and Elimination , respectively.

These are discussed in the following sections, including antibiotic/antifungal therapies and dietary supplements. The Institute for Molecular Medicine is a nonprofit institution and does not endorse commercial products. The products and procedures below are only examples of the types of approaches and substances that could be beneficial to patients with chronic illnesses. Consult your personal physician for advice on treatments, dosing and schedules that can vary for each patient. Antibiotic Therapy for Chronic Chlamydial Infections Patients with Chlamydia species infections or combinations of Chlamydia and Mycoplasma infections appear to do well on long-term combinations of antibiotics plus nutritional and nutraceutical support [1,2]. Examples of antibiotics that have proven useful for treatment of replicating and nonreplicating forms of Chlamydia are discussed here [3]. For replicating, active infections investigators have recommend starting with amoxicillin (initially at 500 mg/day and if tolerated, this dose should be increased to 2X 500 mg/day) [3].
