    Microphthalmia is a relatively rare hereditary eye disorder affecting certain breeds of dogs, including Beagle, Doberman Pinscher, Miniature Schnauzer, Australian Shepherd dog (merle with predominant white color) and other dog breeds. It is characterized by abnormal smallness of the eye.

    Microphthalmia Signs and Causes
    Microphthalmia means the eye(s) started to form during pregnancy but for some reason stopped, leaving the puppy with small eyes. Sometimes, the Microphthalmia condition occurs with coloboma - a structural eye defect when part of the eye does not develop.
    These conditions can affect one or both eyes. Eye affected by microphthalmia can have a very reduced level of vision. With coloboma the visual impairment is variable. When one eye is absent the other eye may be completely normal with good sight, or abnormally developed with poor vision.
    Male dogs are reported to have this condition more often than female dogs.
    The causes of Microphthalmia are considered to be either genetic or environmental, such as toxic substances or diseases (toxoplasmosis or other viral or bacterial infections). They can affect the mother during early pregnancy and damage the fetus.

    General Discussion Lenz Microphthalmia syndrome is an extremely rare inherited disorder characterized by abnormal smallness of one or both eyes (unilateral or bilateral microphthalmos) and/or droopy eyelids (blepharoptosis), resulting in visual impairment. In rare cases, affected infants may exhibit complete absence of the eyes (anophthalmia). Most affected infants also exhibit developmental delay and mental retardation, ranging from mild to severe. Additional physical abnormalities are often associated with this disorder such as an unusually small head (microcephaly) and/or malformations of the teeth, ears, and/or fingers and/or toes (digits). The range and severity of findings may vary from case to case.

    Introduction
    Mutations in the Mitf gene can produce codominant, recessive, or dominant inheritance depending on which part of the protein is affected. Codominant inheritance is produced when the mutation is in DNA binding domain. In this case, the protein dimerizes normally - with protein from normal or mutant allele, but heterodimers can not bind DNA normally (fig.7). With recessive inheritance, the mutations are in the dimerization domains. The mutant protein can not dimerize and therefore cannot bind DNA efficiently (fig. 8). Dominant inheritance patterns have only been found in humans. Here, the DNA binding domain is affected. It has been suggested that human development is more sensitive to haploinsufficiency.
    Materials and Methods
    Single Strand Conformational Polymorphisms (SSCP) and Heteroduplex (HD) analysis of amplified gene segments can effectively screen for single base mutations in nucleic acids. SSCP is based on the fact that single stranded (denatured) DNA molecules take on a sequence specific secondary structure (conformer) when transferred to non-denaturing conditions. Under the same conditions, some of the molecules will renature as a double helix. If there were two DNA sequences, differing only by a single base, heteroduplexes (double helices consisting of one of each sequence) as well as homoduplexes will form. The different conformers of single strands and the different duplexes can be detected due to differing mobilities in gel electrophoresis. used to identify linkage or homology of the merle gene with the mouse or human mitf gene.

    Hi. My daughter was born with microphthalmia in her left eye. At this point, her opthalmologist doesn't think she can see much out of it except for maybe some light or shadows. Her right eye is normal. She started seeing an Ocularist at 4 weeks of age and started wearing a conformer at 5 weeks. The conformer size was increased 3 weeks later and she has been wearing it since then. We have had no problems and she seems to be pretty comfortable with it. She will have her first prosthestic shell when she is closer to 1 year old. I was wondering if anyone out there has gone through a similar situation with there child. If so, did your child's orbit grow as expected with the use of conformers? Are you happy with the appearance of the prosthetic?
    Any advice or information would be greatly appreciated.
