Causes
A person's sex is determined by the X and Y chromosomes. Normally, men have an X and a Y and women have two X's. In Klinefelter syndrome, a male has two X's and a Y. The condition is common and affects 1 in 500 men. The infant appears normal at birth, but the defect usually becomes apparent in puberty when secondary sexual characteristics fail to develop (or develop late). At this time, testicular changes occur that eventually result in infertility in the majority of those affected. Some mild cases may go undetected, with no abnormalities present except infertility. One risk factor for this condition is the mother giving birth at an older age. In most cases, the problem develops during fetal development and is not inherited from one or both parents.

16 Months after Gynecomastia Surgery for Gynecomastia in Klinefelter Syndrome Patient His breast examination revealed mild to moderate breast enlargement with primarily glandular tissue. His testicles were small. His pediatrician assisted with a work up that revealed Klinefelter's syndrome. This is a common cause of gynecomastia and genetic problem seen in males (1:500). Hypogonadism is a problem with small testicles and low hormone output. Some patients do not develop gynecomastia until after puberty.

Charles Plum, a U.S. Naval Academy graduate, was a jet fighter pilot in Vietnam. After 75 combat missions, his plane was destroyed by a surface-to-air missile. Plumb ejected and parachuted into enemy hands. He was captured and spent six years in a Communist prison. He survived that ordeal and now lectures about lessons learned from that experience. One day, when Plumb and his wife were sitting in a restaurant, a man at another table came up and said, "You're Plumb! You flew jet fighters in Vietnam from the aircraft carrier Kitty Hawk. You were shot down!" "How in the world did you know that?" asked Plumb. "I packed your parachute," the man replied. Plumb gasped in surprise and gratitude. The man pumped his hand and said, "I guess it worked!" Plumb assured him, "It sure did if your chute hadn't worked, I wouldn't be here today." Plumb couldn't sleep that night, thinking about that man. Plumb says, "I kept wondering what he might have looked like in a Navy uniform a Dixie cup hat, a bib in the back, and bell bottom trousers. I wondered how many times I might have passed him on the Kitty Hawk. I wondered how many times I might have seen him and not even said 'Good morning, how are you,' or anything because, you see, I was a fighter pilot and he was just a sailor."

Introduction
This booklet was written and published in the light of a tremendous need for further information on Klinefelter's syndrome. It is in no way an attempt of giving a complete description of all aspects of Klinefelter's syndrome. The booklet was written in order to give a better information to Klinefelter-boys and their parents as well as to adult males with Klinefelter's syndrome, but it is also our hope that it will be read by genetic counsellors, physicians, teachers and others who come into contact with boys and adult males with Klinefelter's syndrome. More information on all levels will, however, first and foremost be valuable to Klinefelter-males and their relatives. Publication by the Danish National Society of Turner Contact Groups should be seen as an expression of this society's wish to participate in the spreading of knowledge of Klinefelter's syndrome and create the possibility of establishing Klinefelter contact groups. Furthermore, to make an information, counselling and research center for people with Turner's syndrome and other sex chromosome abnormalities such as Klinefelter's syndrome, triple-X and XYY syndrome. Year after year, from when I was 12 until I was 19, every new doctor said the same thing. "You're Fat. Go on a diet." I could almost read the minds of each one as I walked in their office for the first time. 'Hello Tubby' was written on every scowl and grimace. And year after year, the chants at school of "You need a bra" took me lower and lower down the path of self-defeat. It was a long first year in junior college, and now I was going to go see a new doctor. Here we go again. The doctor looked at me with a concentrated expression. XXY karyotype Klinefelter's syndrome is a condition caused by a chromosome abnormality in males (specifically, a nondisjunction ); affected individuals have a pair of X sex chromosomes instead of just one. It is named after Dr. Harry Klinefelter , the medical researcher who first described this syndrome in Human males usually have an XY karyotype . The XXY chromosome arrangement is one of the most common genetic abnormalities, occurring in about 1 in 1,000 male births, but the symptoms that may result from having the extra chromosome are less common. Because of this extra chromosome, individuals with the abnormality are usually referred to as "XXY Males", or "47,XXY Males" rather than as "suffering from Klinefelter's syndrome." In mammals with more than one X chromosome, the genes on all but one X chromosome are barred from being expressed. This happens in XXY males as well as XX females. A few genes, however, have corresponding genes on the Y chromosome and are not barred. These

Klinefelter's syndrome
Dr Trisha Macnair Klinefelter's syndrome is a chromosomal abnormality that affects males who carry an extra one or more X chromosomes. This can lead to a variety of physical and physiological characteristics.

What are the symptoms? Who is affected? Further help
What are the symptoms?
Features vary from person to person and may not become apparent until puberty. Typical features include tall stature (with disproportionately long legs and arms), pear-shaped fat distribution and delayed puberty. There may be signs of feminisation such as small (or undescended) testes, scant body hair and breast development. Infertility and lack of libido are also common. There may be psychological or behavioural problems concerning self-image. There are also links to thyroid problems, diabetes and osteoporosis.
Who is affected?
The condition is thought to affect between one in 500 and one in 750 births in the UK. Cases are sporadic but there is an increased risk in the children of older mothers. It cannot be cured but testosterone replacement therapy may induce a more male appearance and reduce the risk of osteoporosis in many cases. Older mothers at risk may be offered CVS or amniocentesis pre-natal tests.

Mac users: please note that our site is optimized for the Safari browser Other Klinefelter's syndrome Originally Published: October 26, 2001 Dear Alice, Dear Reader, Men who are born with two X chromosomes [or a genotype (genetic information) of XXY instead of the more typical XY] usually develop a condition called Klinefelter's syndrome. Klinefelter's is the most common sex chromosome disorder, affecting approximately 1 in 500 males across all ethnic and racial groups. Because the "extra" chromosome is a sex chromosome, most of the physical effects of Klinefelter's syndrome become apparent during puberty, if at all. Men with Klinefelter's tend to be tall and have decreased facial hair and muscle mass. They are also more likely to have breast development and very small testicles. Some of these physical differences are thought to be the result of decreased testosterone levels. Injections of synthetic testosterone have been found to increase muscle development and bone density, giving men with Klinefelter's syndrome a more traditionally masculine appearance. However, this replenishment of testosterone cannot correct the infertility that most with Klinefelter's have as a result of the condition. Some research suggests that early (recommended by ages 11 - 13 years) testosterone replacement therapy may decrease the incidence and severity of learning disabilities that can be related to having Klinefelter's, potentially improving overall quality of life. Research on the cognitive problems associated with Klinefelter's are still in the early stages though.

What is Klinefelter syndrome?
Klinefelter syndrome is a genetic disorder that affects males. In Klinefelter syndrome, the male has one or more extra X chromosomes . Males normally have one X and one Y chromosome. Males with Klinefelter syndrome commonly have one extra X chromosome. In rare cases, some males with Klinefelter syndrome have as many as three or four X chromosomes or one or more extra Y chromosomes. Many men with an extra X chromosome are not aware that they have it and lead normal lives. Males with Klinefelter syndrome may be described as XXY males or males with XXY syndrome. Klinefelter syndrome occurs in about 1 in 500 to 1 in 1,000 males.
