On this site, you will find information about X and Y chromosome variations (or aneuploidies), which are commonly known as 47,XXY or Klinefelter syndrome, Triple-X or XXX, 47,XYY and 48-plus X and Y chromosome conditions. In the aggregate, these conditions affect approximately 1 out of every 500 people and translate to approximately 600,000 persons in the U.S. alone. X and Y chromosome variations are common conditions that are rarely diagnosed. And because of that too many individuals do not receive the proven treatments that can significantly diminish symptoms and enhance productivity, physical health and ability to reach life potential. The first of these conditions to be identified was 47,XXY (referring to the fact that the male has 47 chromosomes including 2 X’s and 1 Y). Without proper identification and treatment, persons who are 47,XXY can develop one or more symptoms of Klinefelter syndrome. This web site provides information about when 47,XXY was first discovered, the common characteristics of Klinefelter syndrome, on-going research and treatment. In the near future, we will post information about other X and Y chromosome variations, such as Triple X (referring to females with 47 chromosomes, including 3 X’s) and 47,XYY (referring to males with 47 chromosomes including 1 X and 2 Y chromosomes).

WHAT IS KLINEFELTER SYNDROME? In 1942, Dr. Harry Klinefelter and his coworkers at the Massachusetts General Hospital in Boston published a report about nine men who had enlarged breasts, sparse facial and body hair, small testes, and an inability to produce sperm. By the late 1950s, researchers discovered that men with Klinefelter syndrome, as this group of symptoms came to be called, had an extra sex chromosome, XXY instead of the usual male arrangement, XY. (For a more complete explanation of the role this extra chromosome plays, see the accompanying section, "Chromosomes and Klinefelter syndrome." In the early 1970s, researchers around the world sought to identify males having the extra chromosome by screening large numbers of newborn babies. One of the largest of these studies, sponsored by the National Institute of Child Health and Human Development (NICHD), checked the chromosomes of more than 40,000 infants. Based on these studies, the XXY chromosome arrangement appears to be one of the most common genetic abnormalities known, occurring as frequently as 1 in 500 to 1 in 1,000 male births. Although the syndrome's cause, an extra sex chromosome, is widespread, the syndrome itself-the set of symptoms and characteristics that may result from having the extra chromosome-is uncommon. Many men live out their lives without ever even suspecting that they have an additional chromosome.

Klinefelter's syndrome is found in approximately 1 of 700 men, i.e. there are approximately 3600 boys and men with Klinefelter's syndrome in Denmark with 5 million inhabitants. Men usually have but one X and one Y chromosome, i.e. the chromosomeconstitution 46,XY. Men with Klinefelter's syndrome have more than one X chromosome, usually two X chromosomes, i.e. the chromosome constitution 47,XXY. At birth the testicles of Klinefelter boys are of normal size. When the testicles grow quickly in boys with normal chromosomes at the age of 11-12 years, the testicles of Klinefelter boys stay very small, as a rule only 2 cms. from pole to pole. Usually only few sperms are developed in the testicles, and men with Klinefelter's syndrome are as a rule infertile. Boys with Klinefelter's syndrome should be treated with testosterone, usually from the 11 or 12 years of age, best with Restandol tablets (Testosteroneundecanoat), which does not affect the liver, or with injections of other testosterone preparations. Klinefelter boys more often than other boys have delayed motor function, speech, and maturation development, which does not make them patients demanding special treatment. It is important that these boys at an early age go to a good day-care institution, and that there is a close cooperation between parents and day-care institution staff. If the speech development is delayed, it is important to get help from a speech-therapist for a period. Regarding the motor function development, the participation of parent/child in sports activities and group activities of any kind is of great value. The same is the case with regard to stimulating the delayed development in maturation. In this connection it must be mentioned, that it is also important to stimulate these boys to independence, and not in any way to overprotect them.

A Parent's View
Boys are made of special things and sometimes some Xtra special things! Allow me to begin by telling you that many doctors and researchers do not refer to Klinefelter Syndrome when referring to a child because a child can not and does not have the symptoms of what Harry Klinefelter researched and not all babies and boys will go on to develop the symptoms associated with Klinefelters Syndrome and until they do, they do not have a syndrome. There is the blessing of the early diagnosis. SO much can be changed but just having that advantage. Use it to yours!! So you have just been told that your baby has an extraX.......There is so much more to know than just that! Please take the time to educate yourself. The unknown is a scary place to be and usually the FACTS are not as bad as they first may seem. Please read on!
I was in your shoes just a short time ago when I was told that my two month old baby had this disorder. I was terrified....but I also love my child with all of my heart so I took the time to learn. I will share with you what I felt, heard, read and learned and hopefully provide you with something I wished I had had available to me the day I was told.
