Forebrain development in control and hypothyroid larvae of Triturus cristatus carnifex.
- Gozzo S, Castelvetri IC Int J Neurosci 1988 Feb;38(3-4):299-309. Some cytological and developmental aspects of the forebrain gray matter were studied in the control and the hypothyroid larvae of Triturus cristatus carnifex. The mitotic cell percentages in the periventricular germinal layer, the fusiform shape of the recently divided neuronal cells and the transitory gray matter cleavage, are characteristic of the immature status of the central nervous system (CNS) in this species. The hypothyroidism obtained during two months of thiourea treatment slows down the development in all the forebrain areas of the experimental larvae. Nevertheless, the nucleus olfactorius anterior and the corpus striatum, which have a longer mitotic activity with respect to the other neuronal structures, are more deeply affected by

Comparisons of capillary maturation in control and hypothyroid rat spinal cord: an ultrastructural study.

Quantitative and qualitative features of capillary maturation were examined in the ventral horn of the lumbar spinal cord of control and neonatally induced hypothyroid rats from birth to 6 weeks, using light (LM) and electron microscopy (EM). Quantification of the capillary densities by LM in the control animals and their hypothyroid litter mates have shown three- and twofold increases, respectively, from birth to Postnatal (Pn) Day 21. The following features were observed in the control animals at the EM level: (a) The newborn animals showed varying degrees of capillary maturation; (b) The majority of capillaries possessed mature characteristics by Pn Day 21; (c) By Pn Day 42, mature characteristics were found in nearly all capillary profiles.   • 2 year old female is being evaluated in Peds Endocrinology Clinic for her early-treated congenital hypothyroidism (CH). Is she at increased risk for developmental difficulties even though treated early?
Clinical Bottom Lines
  • Early-treated congenital hypothyroid children do not have increased risk of severe learning disabilities. Subtle deficits are more apparent at early school age and some of these deficits may improve over time. However, with lower school performance found at later school age, this may indicate emerging difficulty with complex skills needed for subjects such as science. Some deficits may be attributable to sensorineural hearing loss associated with poor cochlear development, so hearing screening is important in these patients. Both severity of congenital hypothyroidism at presentation and increased doses of thyroid hormone can put these children at risk for ADD-like problems, necessitating close monitoring and tight control of replacement therapy throughout childhood.

    Development of hypothalamic-pituitary-adrenal response to stress in rats made hypothyroid by exposure to thiouracil from conception

    The functional maturation of the hypothalamic-pituitary-adrenal (HPA) axis has been studied in rats of 20-35 days of age made hypothyroid by the administration of thiouracil from conception. Basal concentrations of corticosterone in serum were normal in hypothyroid animals. Ether stress led to an increase corticosterone content of the adrenal glands of hypothyroid and normal rats but not to a rise in serum corticosterone of hypothyroid rats until 30 days of age. Corticosterone secretion in response to ACTH administration was subnormal in hypothyroid rats. The hypothyroid state delays the development of the hypothalamic-pituitary-portion of the HPA axis until 30 days of age and causes a diminution in adrenal response to ACTH beyond this time.

    Inhibition of Leydig cell activity in vivo and in vitro in hypothyroid rats

    Leydig cell steroidogenic activity under basal and stimulated conditions was studied in hypothyroid rats. Hypothyroidism was induced at a prepubertal age (30 days postpartum) by surgical thyroidectomy, and L-thyroxine (T4) supplementation (6 micrograms/100 g body weight/day for 30 days) to hypothyroid rats was begun after 30 days. Hypothyroidism for 60 days reduced serum LH and FSH without affecting prolactin. Serum and intratesticular testosterone and the specific activity of Leydig cell 3 beta- and 17 beta-hydroxysteroid dehydrogenases diminished in hypothyroid rats. The stimulatory effect of LH on Leydig cell steroidogenic activity and cAMP was also adversely affected in hypothyroid rats. All these changes were reversed by T4 supplementation. The present results suggest that prepubertal hypothyroidism suppresses both basal and stimulated Leydig cell activity in adult rats.

This presentation will describe the case of a 60-year-old woman who,after being hypothyroid for 12 years, developed severe thyroid associated opthalmopathy (TAO) and was found to be hyperthyroid. She had strongly positive anti-TSH receptor antibodies. She developed right optic nerve compression unresponsive to methylprednisolone and requiring orbital decompression at Moorfields Hospital in November 2000. She has subsequently required immunosuppressive therapy for severe TAO affecting the left eye and has received retro-orbital radiotherapy. She is now disabled by diplopia due to enophthalmos of the decompressed eye and muscle tethering of the right eye and is suffering side-effects from steroids. Management of her hyperthyroidism has been complicated by jaundice and an acute arthropathy due to carbimazole and propylthiouracil, and intolerance of beta blockers because of asthma. She underwent total thyroidectomy in March 2001.

Objective: To evaluate the psychometric properties of two new condition-specific questionnaires: (1) the 18-item ThyDQoL, an individualised patient-centred measure of perceived impact of hypothyroidism on QoL, and (2) the 15-item ThySC symptom checklist in which patients provide 'bother' ratings for common hypothyroid symptoms. Method: A cross-sectional survey was conducted with 110 adults with clinical or subclinical hypothyroidism [mean age: 55.1 plus/minus 14.3 (s.d.), range 23-84 years], 103 treated with thyroxine. Respondents rated personally applicable ThyDQoL life domains for importance and impact of their hypothyroidism, and rated ThySC symptoms for bother. Ethical committee approval was obtained. Conclusion: Although measures of hypothyroid symptoms are often and erroneously described as QoL measures, symptoms and QoL are distinct and separate patient-reported outcomes. The new ThyDQoL and ThySC questionnaires will be valuable in routine clinical monitoring and clinical trials of treatments for hypothyroidism.

    The fertility of hypothyroid male mice
    C Chubb and L Henry Male mice homozygous for the hypothyroid gene mutation were compared with normal siblings to determine whether hypothyroidism induced infertility by impairing sexual behaviour. In addition, the fertility of the hypothyroid mice was examined. The experimental results provide unequivocal evidence that genetically-induced hypothyroidism in male mice does not impair sexual behaviour or cause infertility.

    Pituitary response to LHRH in hypothyroid women
    TS Drake, WF O'Brien, and DR Tredway This study was designed to characterize pituitary function in premenopausal women during hypothyroid and euthyroid periods. Six subjects with basal thyroid-stimulating hormone (TSH) levels above 10 microU/ml were studied. Estradiol (E2), prolactin (PRL), follicle-stimulating hormone (FSH), and luteinizing hormone (LH) levels were measured by radioimmunoassay at 30, 15, and minutes before infusion. Pituitary function was evaluated by rapid infusion of 10 micrograms of luteinizing hormone-releasing hormone (LHRH) every 2 hours for a total of 6 hours. Samples were then obtained for FSH and LH every 30 minutes for the duration of the 6-hour study. A significant elevation in basal gonadotropins was observed during the hypothyroid period regardless of basal E2 levels in all 6 subjects (P .05).
