GORR and Ectodermal Dysplasia You’ve probably never heard of a disease called ectodermal dysplasia (ED). It’s a rare disease—thank goodness—but it’s a severe one, a birth defect that negatively affects a victim’s teeth, hair, nails, and even the ability to sweat. Types of ectodermal dysplasia dental problems include unpredictable patterns of tooth eruption and loss, widely spaced teeth, poorly shaped teeth, cavity prone teeth, and the entire absence of teeth. In January 1999 Manhattan resident Susan Barbey wrote to GORR. Her nephew Christopher, a 9-year old living in New York’s suburbs (at the request of the family we will not be using his last name) was born with ED. Christopher has no hair, is unable to sweat, and until very recently had no teeth. But he goes to school, swims competitively, and has even been a YMCA state champion in that sport. He loves his parents, and they love him. So does his aunt.

Ectodermal Dysplasia (ED) is not a single disorder, but a group of closely related conditions of which more than 150 different syndromes have been identified. The Ectodermal Dysplasias (EDs) are genetic disorders affecting the development or function of the teeth, hair, nails and sweat glands. Depending on the particular syndrome ED can also affect the skin, the lens or retina of the eye, parts of the inner ear, the development of fingers and toes, the nerves and other parts of the body. Multiple genes have been discovered to cause ectodermal dysplasias.
Ectodermal dysplasia may affect, to varying degrees, all of the structures associated with the development of the skin. Absent or decreased sweating in people with anhidrotic ectodermal dysplasia is caused by absence of sweat glands. Affected children have difficulty controlling fevers and mild illness may produce extremely high fevers, because of absent temperature regulation by sweating . Affected adults are unable to tolerate a warm environment and require special measures to maintain a normal body temperature
Mucus membrane involvement results in chronic nasal infections with foul-smelling discharge and increased respiratory infections. Hair may be absent or very scanty. The skin is thin with light pigmentation. Teeth develop abnormally and many teeth are missing.

Cattle with anhidrotic ectodermal dysplasia are characterized by phenotypic abnormalities of ectodermal appendages such as hypotrichosis or oligodontia. Affected animals show very sparse hair, reduced number of sweat glands, and missing or defective teeth. We investigated a family of German Holstein cattle with X-chromosomal recessive inheritance of the disease. Comparative analysis of human and murine X-chromosomes revealed a candidate gene for this phenotype called coding for ectodysplasin 1. Mutations in this gene have been shown to be responsible for human anhidrotic ectodermal dysplasia 1 (ED1, also called EDA or XLHED, MIM305100) as well as for the phenotype of the tabby mouse mutant.
