Definition of Arteriohepatic Dysplasia
Arteriohepatic dysplasia is a genetic disorder that mimics other forms of prolonged liver disease in infants and young children. However, a group of unusual features in other organ systems distinguishes Arteriohepatic dysplasia from other liver and bile duct diseases in infants. It is also known as Allagile syndrome. The gene for this disorder has been discovered on chromosome 20 in band 20p12.

Definition of Arteriohepatic Dysplasia
Arteriohepatic dysplasia is a genetic disorder that mimics other forms of prolonged liver disease in infants and young children. However, a group of unusual features in other organ systems distinguishes Arteriohepatic dysplasia from other liver and bile duct diseases in infants. It is also known as Allagile syndrome. The gene for this disorder has been discovered on chromosome 20 in band 20p12.
Description of Arteriohepatic Dysplasia
Children with arteriohepatic dysplasia usually have a liver disease characterized by a progressive loss of the bile ducts within the liver over the first year of life and narrowing of the bile ducts outside the liver. This leads to a buildup of bile in the liver, causing damage to liver cells. Scarring may occur and lead to cirrhosis in about 30 to 50 percent of affected children. Arteriohepatic dysplasia was first described in 1969 by D. Alagille, in French medical literature.

It is an idiopathic disorder characterised by cholestatic jaundice, hepatomegaly and often splenomegaly. There is a nonsyndromic form of intrahepatic bile duct hypoplasia without the extrahepatic features. The natural history is for progressive liver damage leading to death in the second or third decades. In the syndromic type (see Alagille syndrome ) vertebral body abnormalities, which include butterfly and hemivertebra are present characteristically in the thoracic spine. Hepatic sonography is usually normal though a gallbladder may not be visible. There is no intrahepatic bile duct dilatation. Hepatobiliary scintigraphy reveals similar findings to that in extrahepatic biliary atresia, i.e. normal or slightly impaired hepatic uptake and no evidence of excretion of tracer into bile ducts or bowel. Differentiation from extrahepatic biliary atresia requires liver biopsy and/or cholangiography. See atresia biliary

The Transplant Center is a partnership of the following entities: Our liver transplant program is one of the world's oldest Liver Disorders Transplant Services Website Arteriohepatic Dysplasia (Alagille Syndrome) Alagille syndrome is a genetic condition affecting 1 in 70,000 newborns. Generally presenting in infancy, this condition is often misdiagnosed as biliary atresia. Occasionally, it can escape medical attention until school age, where it appears as xanthomata (fibrous, fatty tissue or nodules under the skin) or pruritis (itching). Jaundice may or may not be present. The syndrome is marked by five major features, several of which must be present for diagnosis: cholestatic liver disease, congenital heart disease, vertebral and/or long bone anomalies, ophthalmic (eye) abnormalities and peculiar facies (facial features), such as deep-set eyes and an oversized brow. There is a 50 percent chance that people with this syndrome will pass it to their children. A prenatal diagnosis is not available.
