An innovative approach to recent developments Published online first All articles are published electronically ahead of print with the definite citation line. Thus, an article will be available online shortly after the author's approval to print. Aims and Scope In the past decade research into skin pharmacology has developed dramatically with new and promising drugs and therapeutic concepts being introduced regularly. Recent examples include immunosuppressive topicals, and new strategies in the therapy of malignant melanoma and wound healing. New concepts have also been developed in traditional areas of skin therapy and skin care which have led to discussions of issues such as ‘cosmeceuticals’ in the United States, ‘quasi-drugs’ in Japan, and the 6th Amendment of the Cosmetic Regulation in the European Union. There are many open questions which reflect the importance of the subject and the need for a new international scientific forum where they can be discussed in extenso. Dermatologists, pharmaceutical chemists, pharmacologists, toxicologists, and others interested in the subject are invited to contribute to this exciting project and to submit their best research work for publication. ABSTRACT
B ACKGROUND: Although opioid analgesics have well-defined efficacy and safety in treatment of chronic cancer pain, further research is needed to define their role in treatment of chronic noncancer pain. OBJECTIVE: To evaluate the effects of controlled-release oxycodone (OxyContin tablets) treatment on pain and function and its safety vs placebo and in long-term use in patients with moderate to severe osteoarthritis pain. METHODS: One hundred thirty-three patients experiencing persistent osteoarthritis-related pain for at least 1 month were randomized to double-blind treatment with placebo (n = 45) or 10 mg (n = 44) or 20 mg (n = 44) of controlled-release oxycodone every 12 hours for 14 days. One hundred six patients enrolled in an open-label, 6-month extension trial; treatment for an additional 12 months was optional. RESULTS: Use of controlled-release oxycodone, 20 mg, was superior (P <.05) to placebo in reducing pain intensity and the interference of pain with mood, sleep, and enjoyment of life. During long-term treatment, the mean dose remained stable at approximately 40 mg/d after titration, and pain intensity was stable. Fifty-eight patients completed 6 months of treatment, 41 completed 12 months, and 15 completed 18 months. Common opioid side effects were reported, several of which decreased in duration as therapy continued. CONCLUSIONS: Around-the-clock controlled-release oxycodone therapy seemed to be effective and safe for patients with chronic, moderate to severe, osteo-arthritis-related pain. Effective analgesia was accompanied by a reduction in the interference of pain with mood, sleep, and enjoyment of life. Analgesia was maintained during long-term treatment, and the daily dose remained stable after titration. Typical opioid side effects were reported during short- and long-term therapy.

    Pharmacology and Toxicology at Queen's University
    The Department of Pharmacology and Toxicology began life in 1854 with the appointment of Materia Medica. The initial responsibility was primarily the education of medical students which was shouldered by a number of appointees until 1938 when Dr. Eldon Boyd was appointed Professor and Head of the Department of Pharmacology. With his appointment, the training of graduate students was initiated; some 26 theses were completed during the ensuing 30 years. With the appointment in 1969 of Dr. Gerald S. Marks, the department began a period of rejuvenation with the hiring of several new faculty members, a change in its research profile, and new forays in teaching with participation in the instruction of Arts and Science undergraduates through the nascent Life Sciences program in 1970. Prior to 1969, the teaching responsibilities were limited to 75 medical students and a handful of graduate students. From 1970 to 2000, 176 graduate students completed their theses.
    In 2003/2004, the department comprised ten faculty members, four and a half support staff, and 40 graduate students. The primary undergraduate teaching responsibilities were 80 medical students, 70 nursing students, and the rapidly growing Life Sciences program.

    SSRI. Symptomatic relief of depressive illness: Initially, usually in the morning, a 25 mg single daily dose. Patients unresponsive to a 25 mg dose may benefit from dose increases, in 12.5 mg/day increments, at intervals of at least 1 week, to a maximum of 62.5 mg/day. Symptomatic relief of panic disorder: Initially, usually in the morning, a 12.5 mg single daily dose. Dose changes at intervals of at least 1 week should occur in 12.5 mg/day increments. Maximum dosage: 75 mg/day. Social phobia (social anxiety disorder): Initially, usually in the morning, a 12.5 mg single daily dose.     Antipsychotic Agent Loxapine, a tricyclic dibenzoxazepine antipsychotic agent, which is chemically distinct from the phenothiazines, thioxanthenes and butyrophenones, produces pharmacological responses in various animal species which are characteristic of those seen with the majority of antipsychotic drugs. Although the precise mechanism of action is not known, loxapine succinate administration results in strong inhibition of spontaneous motor activity. Absorption following orally administered loxapine succinate is virtually complete. Following oral administration of a single 25 mg dose of loxapine, the onset of sedative effect occurs in 15 to 30 minutes; peak effect occurs within 1.0 to 3 hours. The duration of sedative effect is approximately 12 hours. Loxapine is metabolized rapidly and extensively in animals and man and excreted both in the urine and feces. There appear to be 3 reactions, other than phenol conjugation, involved in the metabolism of loxapine: aromatic hydroxylation, N-oxidation and N-demethylation. Unmetabolized drug has not been recovered in urine and feces though metabolites are excreted in urine mainly in the form of conjugates and in feces mainly in the unconjugated form. In rats, 86 to 96% of orally administered doses of loxapine has been recovered in urine and feces. In dogs, 92 to 104% has been similarly recovered. Approximately 50% of the drug is recovered within the first 24 hours.
