General Microbiology
GENERAL Bacteria are unicellular microorganisms having a variety of characteristics allowing their classification. One major classification scheme is based upon their staining properties using the "Gram stain" procedure. In this procedure, heat-killed bacteria are exposed to the purple dye crystal violet and iodine. This combination forms a dye complex in the bacterial cell wall. Treatment of the stained bacteria with a decolorizer like ethanol will wash away the dye complex from some bacteria but not others. Bacteria that retain the crystal violet-iodine complex appear purple and are called "Gram-positive". Bacteria that lose the dye complex can be counterstained with the red dye saffranin so that they appear red. These bacteria are called "Gram-negative". The basis of the Gram reaction lies within the structure of the cell wall, described below. Bacteria also come in many different shapes. Spherical shapes are referred to as "cocci" while elongated cylinders are called "bacilli" or "rods". Some bacteria are slightly elongated cocci and these are referred to as "coccobacilli". Even other bacteria have a corkscrew-like appearance; these spiral forms are often called "spirochetes". Individual cells may also be arranged in pairs or clusters or chains. Thus, may morphologies are possible and these can be useful for the identification of bacterial genera. ( Click here to see a bacterial classification flowchart The ability of a bacterium to cause disease is known as its virulence. Factors involved in determining virulence potential are discussed

A note about your final grades
Your final grades were calculated and verified by me and the two laboratory instructors. In the unlikely event that you discover a mistake in the computation of your final grade, a grade change will be considered. It is futile to contact me for explanations of "curves" and for discussions of personal problems that may have affected your performance. In a class having more than 80 students, there are invariably a number of misfortunes, including students who are victims of accidents or who learn that they (or members of their family) have cancers, require major surgery, etc. In cases involving serious disruptions, a request of an "incomplete" along with proof of the cause of the disruption, well before the end of the quarter, is advisable. A grade is earned, not granted. It certifies a level of understanding of the subject matter. It bears little relationship to the performance of the other students, and it has no relationship whatsoever to a student's personal problems or to his/her future plans. Anything else would devalue the reputation of the University.

How Germs Can Zap You and How You Can Zap Back , Warner Books, Publisher, publication date, October 1, 2001 was written by me and Edited by Dinah Dunn of Byron Preiss Visual Publications, New York and Jackie Merri Meyer of Warner Books. Available on-line and at bookstores, the book provides information on microorganisms associated with foodborne illness, colds and flu, day care, hospitals, travel, pets, home, work and those that may be used as an agent for biological warfare.
All of the "Feature Articles," and the articles in "What the Heck is...?" and in "General Interest" were written by me, and therefore, any mistakes are mine, alone. I have tried to be as accurate as possible within the limits of providing the information in a "reader-friendly" format. Therefore, please forgive any latitude I have taken with the pure science discussed. With these caveats in mind: in keeping with the spirit of the "Web" and Internet, and the fact that this institution has been established for, and is devoted to, learning, all of the articles on these Pages are for anyone's use, as long as the use is for non-profit only, and this statement accompanies any copies.

39. General Immunology
Section of Introduction to Medical microbiology course provides understandable discussion of basic immunological concepts and their relationship to infectious disease from Southern Illinois Univ.
http://www.cehs.siu.edu/fix/medmicro/genimm.htm
General Immunology
Host defense is present in many forms. Overall, the Immune Response (IR) can be divided into two major classifications; humoral and cell-mediated. While these responses are not mutually exclusive, they provide distinctly different avenues for dealing with pathogenic organisms or altered host cells. These different responses will be discussed in more detail later. Immune Response Humoral Immunity Cell-mediated Immunity (Antibody) (Cytotoxicity) Some of these responses are specific, others are non-specific. This page will introduce host defense mechanisms by defining some commonly used terms and describing the specific cells and tissues involved in these immune responses. DEFINITIONS: Antigen (Ag): A molecule which elicits a specific immune response when introduced into an animal. More specifically, antigenic (immunogenic) substances are:
  • Structurally complex (proteins are usually very antigenic),
  • Accessible (the immune system must be able to contact the molecule), and
  • Foreign (not recognizable as "self"). Antibody (Ab): A glycoprotein produced in response to an antigen that is specific for the antigen and binds to it via non-covalent interactions. The term "immunoglobulin" is often used interchangeably with "antibody". We will use the term "immunoglobulin" to describe
