Abstract
The genetic basis of large adaptive differences in timing of bud set and frost hardiness between natural populations of Scots pine ( Pinus sylvestris L .) was studied with the aid of RAPD markers and quantitative genetic tools. Steep clinal variation was found for both traits among Finnish Scots pine populations, and the differences between populations were found to be largely genetic. QTL mapping with Bayesian analysis revealed four potential QTLs for timing of bud set, and seven for frost hardiness. The QTLs were mostly different between the two traits. The potential QTLs included loci with large effects, and additionally smaller QTLs. The largest QTLs for bud set date accounted for about a fourth of the mean difference between populations. Thus, natural selection during adaptation has resulted in fixation of genes of large effect. This result is in conflict with the classical infinitesimal model, but agrees with the results of Orr (1998), suggesting fixation of large effects during adaptation.

Abstract
The nature of symptoms and signs of sciatica, genetic factors, and efficacy of periradicular infiltration were studied in 160 nonoperated patients with unilateral sciatica of 3 to 28 weeks duration. Back and leg pain (100-mm VAS), disability (Oswestry), and quality-of-life (NHP) were evaluated. ENMG and 1.5-T MRI were performed on every patient. Presence of the Trp2 and Trp3 alleles of collagen IX was determined from blood samples. After informed consent, patients were randomized for periradicular infiltration with either methylprednisolone-bupivacaine, or saline. The final follow-up assessment was 1 year after the intervention. Economic analysis was based on data gathered from the patients, medical records and the National Insurance Register. Periradicular infiltration with methylprednisolone-bupivacaine produced a significant treatment effect compared to saline at 2 weeks for leg pain, straight leg raising, lumbar flexion and patient satisfaction. At 6 months, saline was superior to steroid in back and leg pain. By 1 year, 18 patients in the methylprednisolone group and 15 in the saline group had received surgical treatment.

Current Research Program My research focuses on the large brown algae, the kelp (Laminariales). Presently, our laboratory is concentrating on two aspects of kelp biology: 1. evolution and population genetic diversity and 2. marine agronomy. Evolution and genetic diversity are being explored using molecular tools. Our contributions are most advanced in our evolution studies. We have examined chloroplast and ribosomal DNA diversity among kelp species, genera and families, and between the kelp order Laminariales, other orders of brown algae, and other divisions of plants. These studies have provided provocative insights into kelp evolution; for example: 1. We were able to demonstrate a surprisingly close evolutionary relationship of kelp and the freshwater fungus Achlya . 2. On the basis of ribosomal and chloroplast DNA data we have proposed that the taxonomy of the Laminariales is greatly inflated and suggested a reduction of advanced families of kelp from three to one, a reduction of kelp genera from 25 to five, and a reduction of kelp species from well over 100 to 25. 3. Application of a molecular clock to our DNA data suggests kelp genetically diverged 16-22 million years ago, thus, evolving simultaneously with the modern terrestrial legumes. Earlier estimates, based on questionable fossil fragments, had these 3dinosaurs of the plant world2 diverging very much earlier.

Parque Nacional Soberania, Panama Our lab carries out studies in the areas of evolutionary biology, genetics and molecular systematics. Our principal research interest is biogeography and history-based analysis of community assembly, diversification and maintenance. We have focused our attention on two regional community assemblages:1) the birds of the Lesser Antilles the neotropical freshwater fishes of lower Central America . The lab uses the tools of molecular biology to infer the phylogenetic and demographic histories of the taxa comprising these avian and freshwater fish communities. Because we are interested in the tempo as well as the mode of evolutionary change in these communities, we also study the behavior of molecular clocks. Our molecular clock studies have tended to focus on the molecular divergence between marine species pairs found on either side of the isthmus of Panama. Our lab is part of the Naos Laboratories located near the Pacific entrance of the Panama Canal in the Republic of Panama. The lab is part of STRI's Molecular Systematics and Evolution Group along with the labs of Haris Lessios, Nancy Knowlton and a visiting scientists laboratory.
