Feb. 9, 1994, Madison, Wis.), American virologist who in 1975 shared the Nobel Prize for Physiology or Medicine with his former professor Renato Dulbecco and another of Dulbecco's students, David Baltimore , for his codiscovery of the enzyme reverse transcriptase. While working toward his Ph.D. under Dulbecco at the California Institute of Technology, Temin began investigating how the Rous sarcoma virus causes animal cancers. One puzzling observation was that the virus, the essential component of which is ribonucleic acid (RNA), could not infect the cell if the synthesis of deoxyribonucleic acid (DNA) was stopped. Temin proposed in 1964 that the virus somehow translated its RNA into DNA, which then redirected the reproductive activity of the cell, transforming it into a cancer cell. The cell would reproduce this DNA along with its own DNA, producing more cancer cells. Skeptics pointed out that Temin's suggestion contradicted the contemporary tenet of molecular biology: that genetic information always passed from DNA to RNA, rather than the reverse. But in 1970 both Temin and Baltimore proved Temin's hypothesis correct. They identified an enzyme (reverse transcriptase) in the virus that synthesizes DNA that contains the information in the viral RNA. Temin obtained his Ph.D. in 1959, and after spending another year with Dulbecco, he joined the faculty of the University of Wisconsin in Madison, where he taught and conducted research until his death.

Temin was born in Philadelphia and educated at Swarthmore College, Pennsylvania, and the California Institute of Technology. From 1960 he worked at the University of Wisconsin, becoming professor 1969.
Temin's prizewinning research was on a virus that has a mechanism which incorporates its material into mammalian genes. He discovered that beneficial mutations outside the germ line are naturally selected and that the mechanism adds genetic information from outside into the germ line.

Article in a Box Howard Martin Temin – Co-discoverer of Reverse Biological Information Flow S Mahadevan One of the first observations of Temin, working under the guidance of Harry Rubin, a post doc in Dulbecco's lab, was that when RSV was added to a sparse layer of chicken embryonic cells, infected centers showed up as foci, the cell-culture equivalents of tumours. Even today, this is an important assay for the tumerogenic potential of a virus. Temin also found that the morphology of the foci were different when genetic variants of the virus was used, leading to the important conclusion that transformation is the result of action by genes carried by the virus. Subsequent studies by others established that these are cellular genes `hijacked' by the virus, whose function has been modified. Thus it was realized that modification of normal cellular genes can lead to cancer. the viral RNA genome is used as a template to make a DNA copy that is integrated into the host genome. The DNA provirus is then used as a template that is transcribed to give the viral RNA copies.

In groundbreaking work in the late 1950s, he and his colleagues demonstrated that, starting only with the hydrogen and helium produced in the Big Bang, all the elements from carbon to uranium could be produced by the nuclear processes in stars.

He won a Nobel Prize in Physiology or Medicine in for describing how tumor viruses act on the genetical material of the cell through reverse transcriptase. This upset the widely held belief at the time of the "Central Dogma" of molecular biology posited by Nobel laureate Francis Crick , one of the co-discoverers of the structure of DNA (along with James Watson and Rosalind Franklin ). Crick, along with most other molecular biologists of the day, believed genetic information to flow exclusively from DNA to RNA to protein . Temin showed that certain tumor viruses carried the enzymatic ability to reverse the flow of information from RNA back to DNA using reverse transcriptase. This phenomenon was also independently studied by David Baltimore , with whom Temin shared the Nobel Prize. The discovery of reverse transcriptase is one of the most important of the modern era of medicine, as reverse transcriptase is the central enzyme in several widespread human diseases, such as HIV , the virus that causes AIDS , and Hepatitis B . Reverse transcriptase is also an important component of several important techniques in molecular biology and diagnostic medicine, such as the polymerase chain reaction (PCR).
