Frederick Sanger received two Nobel prizes (in the same category), for his work on protein sequencing and DNA sequencing.
FREDERICK SANGER Frederick Sanger was born in Rendcombe, England. His father was a medical doctor and it was expected that Fred would also enter the medical field. As Sanger grew up, he became very interested in nature and science and when he went to Cambridge University , he made the decision not to study medicine. He felt that a career in science would give him a better chance to become a problem solver. Sanger was a conscientious objector during the war because of his Quaker upbringing. After his B.A. in 1939, he stayed at Cambridge to do a Ph.D. with Albert Neuberger, on amino acid metabolism. After his Ph.D. in 1943, Sanger started working for A. C. Chibnall, on identifying the free amino groups in insulin. In the course of identifying the amino groups, Sanger figured out ways to order the amino acids. He was the first person to obtain a protein sequence. By doing so, Sanger proved that proteins were ordered molecules and by analogy, the genes and DNA that make these proteins should have an order or sequence as well. Sanger won his first Nobel Prize for Chemistry in 1958 for his work on the structure of protein.

    SELECTED PAPERS OF FREDERICK SANGER
edited by (Retired from Medical Res. Council Lab. of Molecular Bio., Cambridge)
This important volume is mainly concerned with the development of methods for "sequencing" — that is, determination of the order of the amino acids in proteins and of nucleotides in RNA and DNA. In 1943 the position of only one amino acid in a protein (insulin) was known, and Sanger's first paper resulted in finding a second amino acid. In his final paper in 1982 he describes the determination of a DNA sequence of 48,502 nucleotides. The papers describe the steady improvements in techniques, and exciting biological results revealed by the sequences.

Sanger determined the complete amino acid sequence of insulin . In doing so, he proved that proteins have specific structures. He began by degrading insulin into short fragments by mixing the trypsin enzyme (which splits protein) with an insulin solution . He then applied a spot of the mixture to a sheet of filter paper. He passed a solvent through the filter paper in one direction, and passed an electric current through the paper in the opposite direction. Frederick Sanger (b. 1918). British Chemist and Two-time Nobel Laureate (1958, 1980). Very Interesting Autograph Letter Signed, one page, octavo, no date, but envelope of transmission postmarked 1996, Cambridge, England. Reads in part: "I don't think there is any absolute obligation to attend the Nobel celebrations" and that in 1958 when he was in Stockholm the "only winner who was not there was Boris Pasternak who was forbidden by the Russian authorities to come. Yours sincerely, Fred Sanger".
Sanger won Nobel in 1958 for studies on insulin and in 1980 for research on nucleic acids. A very fine letter, with good relational content, shows normal aging and handling, cross-folds, accompanied with original envelope of transmission. $OLD
