Prion Proof? Evidence grows for mad cow protein
Nathan Seppa Prusiner, a neurologist and biochemist at the University of California, San Francisco (UCSF), and his colleagues now report results that could silence many of his critics. The study, published in the July 30 Science , shows that purified prions can cause disease when injected into the brains of genetically engineered mice. Previous work by Prusiner and others had implicated prions in human-brain ailments that include Creutzfeldt-Jakob disease and kuru, as well as mad cow disease, chronic wasting disease in deer and elk, and scrapie in sheep. Suggesting that proteins, misfolded or not, can be infectious is "a radical notion," says Neil R. Cashman of the University of Toronto. Nevertheless, to Cashman and others, the new research supplies the proof. Previous animal experiments with infectious preparations of natural prions hadn't provided strong evidence of infectivity. Nucleic acids, such as viral DNA or RNA, might have tainted the preparations, says Chih-Yen King of Florida State University in Tallahassee.

Prusiner discovered and characterized an entirely new class of proteins called prions, which cause several rare and fatal neurodegenerative diseases. A Nobel Prize for Alumnus Stanley Prusiner The history of Penn winners of the Nobel Prize in Medicine starts with Dr. Gerald Edelman (M '54), who shared the prize in 1972 for his work on the chemical structure of antibodies. Next came the 1976 Prize to Dr. Baruch Blumberg (who was on the SAS faculty in anthropology at the time) for his discovery of the antigen involved in Hepatitis B. In 1985 the dual alumnus Dr. Michael Brown (C '62, M '66) shared the Prize for his work in cholesterol and metabolic diseases. That same year, a group of medical faculty including Dr. Patrick Storey shared in the Peace Prize. In Dr. Edelman's year the 1972 Chemistry Prize was also shared by another alumnus whose work contributed to medicine-particularly blood diseases- Dr. Christian Anfinsen, who had taken his master's degree here. This year's winner of the Nobel Prize in Medicine is Dr. Stanley B. Prusiner (right), a Penn alumnus who was "all by himself against the world" for many years in his pursuit of the prion, a new class of pathogen that is now accepted as the infectious agent in "mad cow disease" and in human neurodegenerative diseases such as Creutzfeldt-Jakob disease. "

On Oct. 23, Prusiner Will Speak about His Research on Toxic Proteins
Stanley Prusiner olumbia will award the 1997 Louisa Gross Horwitz Prize to Stanley B. Prusiner, professor of neurology and biochemistry at U.C.-San Francisco this Thursday (Oct. 23). Prusiner, winner of this year's Nobel Prize in Medicine, is to be honored for his groundbreaking work on prions, a protein capable of transmitting degenerative neurological diseases. The Horwitz Prize, established in 1967, is given annually for outstanding research in biology or biochemistry. Research previously recognized by other awards, such as the Nobel Prize, is not eligible. The Horwitz Prize committee selected Prusiner for the award in May. They have been linked to certain neurodegenerative disorders, such as bovine spongioform encephalitis (better known as "mad cow disease") and Creutzfeldt-Jakob disease, which is similar to mad cow disease but occurs in humans. In these diseases, holes develop in the brain, resulting in dementia and, eventually, death. In England, more than 130,000 cows have been affected by mad cow disease since the 1980s. Scientists believe the cows were infected by a food supplement made of the meat and bone meal from sheep that had scrapi. Researchers are now investigating whether people who ate the contaminated beef subsequently developed mad cow disease.

Prusiner, M.D., recipient of the 1997 Nobel Prize in Physiology or Medicine, will deliver a lecture at 4 p.m. Thursday, April 6 at the University of Iowa Levitt Center for University Advancement. Prusiner's lecture, "Prions and the Brain," is a regional event sponsored by the Institute of Medicine of the National Academy of Sciences. Prusiner is director of the Institute for Neurodegenerative Diseases and professor of neurology and biochemistry at the University of California, San Francisco (UCSF). An editor of eight books and author of more than 250 research articles, Prusiner's contributions to scientific research have been internationally recognized. His Nobel Prize-winning research centered on his discovery of prions, protein particles that are considered to be the cause of various infectious diseases of the central nervous system, such as scrapie, "mad cow disease" and Creutzfeldt-Jakob disease. Prusiner proposed what many scientists considered to be the heretical idea that prions are composed only of protein and are devoid of nucleic acid. Previously, scientists believed that transmissible central nervous system diseases were caused by slow-acting viruses. Prusiner's studies demonstrated how an infectious pathogen lacking nucleic acid could multiply and cause central nervous system degeneration. His work significantly changed the way scientists and physicians think about diseases of the central nervous system and may have profound implications for future biomedical research.

Prusiner kom med prionteorien tidlig på 1980-tallet møtte han stor motstand i de vitenskaplige miljøene. Men Prusiners forskning har vist at prionteorien er høyst aktuell og reell. I 1997 fikk Prusiner Nobelprisen i medisin for sin prionforskning. Prionteorien Prioner er smittestoff som kun består av proteiner og som ikke er avhengig av arvestoff slik som alle andre kjente smittestoff, herunder virus og bakterier.

FRANKFURTER RUNDSCHAU Den folgenden Artikel fanden wir in der Frankfurter Rundschau vom 11.10.1997 ( er wurde etwas vereinfacht ) Gegen heftige Anfeindungen der Infektionsbiologen Der Medizin-Nobelpreis geht an den US-Neurologen Stanley B. Prusiner Von Harald Schlatter wissenschaftliche Auszeichnung schon jetzt zuteil wurde - denn bislang ist seine bahnbrechende bewiesen. Ausgangspunkt der Studien Prusiners war eine Patientin, die 1972 an der Creutzfeldt-Jakob-Krankheit (CJK) verstarb. Neben einer spontanen und einer vererbbaren Form dieser Hirnschwammerkrankung existiert auch eine dritte, eine Bereits mehrfach ist es durch unzureichend Erregerhindeutet. Die Suche nach ihm blieb aber erfolglos. Das hatte CJK mit Scrapie gemein, einer Hirnschwammerkrankung, die seit mehr als 200 Jahren vorwiegend unter Schafen grassiert. Scrapie : Wiesen, auf denen eine infizierte Herde geweidet hatte, mieden sie mehrere Jahre. Die besorgniserregende Resistenz des damals oder mit ionisierender Strahlung, die normalerweise das Erbgut absterben, konnten dem Hirnschwammerreger nichts anhaben.
