Otto Loewi was born in 1873 in Frankfort, Germany. A pharmacologist, he investigated how vital organs respond to chemical and electrical stimulation, and established their dependence on certain proteins for proper function. As a result, he learned how nerve impulses are transmitted by chemical messengers. His findings helped advance medical therapy and earned him the 1936 Nobel Prize for Physiology or Medicine. Otto Loewi
Born in Frankfurt am Main, Germany, Otto Loewi originally wanted to be an art historian but bowed to family pressures and went to medical school instead (similarly, Ramón y Cajal's plans to become a painter were thwarted by his father). Loewi later went into pharmacology research and was one of the first people to propose that cells talked to one another with "little whiffs of scent" - more formally to be known as the chemical theory of synaptic transmission. While living in Graz, Austria in 1921, Loewi dreamed of an experiment that would win him the Nobel Prize - "The night before Easter Sunday of that year I awoke, turned on the light, and jotted down a few notes on a tiny slip of thin paper. Then I felt asleep again. It occurred to me at six o'clock in the morning that I had written down something most important, but I was unable to decipher the scrawl. The next night, at three o'clock, the idea returned. It was the experiment to determine whether or not the hypothesis of chemical transmission that I had uttered seventeen years ago was correct. I got up immediately, went to the laboratory, and performed a simple experiment on a frog heart according to the nocturnal design" Excerpt from Otto Loewi: An Autobiographic Sketch (Univ. of Chicago Press, 1960)

Otto Loewi
Nobel laureate from the University of Graz
The Nobel laureate Otto Loewi worked and conducted research for decades at the University of Graz. Being of Jewish descent, however, he was forced to leave the country in 1938 and emigrated to the USA. Otto Loewi is born in Frankfurt am Main, Germany. The medical doctor qualifies as a university lecturer in Marburg (Germany). His research proves that, contrary to what was believed at the time, the animal organism is capable of synthesising proteins from amino acids. The German citizen Loewi accompanies his professor from Marburg to Vienna to assist in his researches there. In 1909, Loewi - who has by now become an Austrian citizen - goes to Graz, where he accepts a chair of pharmacology at the University there. In the years 1912/13 he also serves as dean of the faculty of medicine. In 1936, Otto Loewi receives the Nobel Prize for his outstanding research into the transmission of nerve impulses to organs. The famous scientist is forced to leave Austria because of his Jewish origins.

Otto Loewi's Frog Heart Experiments The Problem: In 1903 Otto Loewi wondered why one of the heart nerves (the accelerator) speeds up the heart and the other (the vagus) slows it down, even though the electrical pulses of the nerves were nearly identical. The Hypothesis: Loewi suggested that the 2 nerves released different chemicals at their terminals when stimulated. The vagus chemical would slow down the heart and the accelerator chemical would speed it up. The Experimental Test Seventeen years later Loewi had a very strange dream in which a way of testing his hypothesis appeared. He woke up and wrote down some notes, but the next morning he could not read his scribbles! Luckily for him the dream reoccurred the next night and he immediately got up (3AM) and did the experiment. Hearts, with their nerves intact, were dissected from 2 frogs. The hearts were hooked up to mechanical devices which recorded their heart beats. Then the vagus nerve of one heart (the donor) was electrically stimulated. The donor heart immediately slowed down, as expected. A sample of the fluid passing through the donor heart was taken with a small pipette and this fluid was dripped onto the second heart (the recipient). The recipient heart slowed down, even though its vagus had not been stimulated. Chemicals released by the donor heart into the fluid were sufficient to produce slowing. (Note: the picture shows one of several ways of doing this experiment. In some of the early experiments the fluid from the donor heart was collected in a beaker and the recipient heart was placed directly into the beaker).

He used this preparation almost throughout his whole career, and it was this preparation that he used for the experiments which lead to his Nobel Prize. Inactivation

Communication of information between neurons is accomplished by movement of chemicals across a small gap called the synapse . Chemicals, called neurotransmitters , are released from one neuron at the presynaptic nerve terminal. Neurotransmitters then cross the synapse where they may be accepted by the next neuron at a specialized site called a receptor. The action that follows activation of a receptor site may be either depolarization (an excitatory postsynaptic potential) or hyperpolarization (an inhibitory postsynaptic potential). A depolarization makes it MORE likely that an action potential will fire; a hyperpolarization makes it LESS likely that an action potential will fire.
    Discovery of Neurotransmitters
Back in 1921, an Austrian scientist named Otto Loewi discovered the first neurotransmitter. In his experiment (which came to him in a dream), he used two frog hearts. One heart (heart #1) was still connected to the vagus nerve. Heart #1 was placed in a chamber that was filled with saline. This chamber was connected to a second chamber that contained heart #2. So, fluid from chamber #1 was allowed to flow into chamber #2. Electrical stimulation of the vagus nerve (which was attached to heart #1) caused heart #1 to slow down . Loewi also observed that after a delay
