Signal¶verf¶ringen sker i speciella kontaktpunkter, synapser . En enda nervcell kan ha tusentals kontaktpunkter med andra nervceller. Carlsson har uppt¤ckt att dopamin ¤r en s¥dan signalsubstans i hj¤rnan och att dopamin har stor betydelse f¶r kontroll av v¥ra r¶relser. Hans forskningsr¶n ledde i sin tur till insikten att Parkinsons sjukdom orsakas av dopaminbrist i vissa delar av hj¤rnan och till att man kunde f¥ fram ett effektivt l¤kemedel (L-dopa) mot denna sjukdom. Greengard uppt¤ckte hur dopamin och en rad andra signalsubstanser ut¶var sina effekter i nervsystemet. Signalsubstanserna p¥verkar f¶rst en receptor p¥ cellens yta. Det utl¶ser en kaskad av reaktioner som p¥verkar vissa "nyckelproteiner" som i sin tur reglerar olika funktioner i cellen. Proteinernas form och funktion f¶r¤ndras genom att fosfatgrupper tillf¶rs (fosforylering) eller tas bort (defosforylering). Genom denna mekanism kan signalsubstanser ¶verf¶ra sina budskap mellan nervceller. Kandel uppt¤ckte sedan hur synapsernas effektivitet kan f¶r¤ndras och med vilka molekyl¤ra mekanismer detta sker. Han visade att synapsernas funktion ¤r central f¶r inl¤rning och minne. F¶r uppkomsten av en form av korttidsminne spelar proteinfosforylering i synapsen en viktig roll. F¶r att ett l¥ngtidsminne ska uppst¥ kr¤vs dessutom nybildning av proteiner som bl.a. leder till att synapsens form och funktion f¶r¤ndras.

Summary: Eric Kandel's lab is studying examples of the major forms of memory storage. The lab is studying explicit memory storage (the conscious recall of information about people, places, and objects) in mice and implicit memory storage (the unconscious recall of perceptual and motor skills) in the snail Aplysia . In Aplysia , the lab has focused on the implicit memory for sensitization, a simple form of learned fear, and the mechanisms for achieving synapse-specific anatomical changes. In mice, they have examined the synaptic mechanisms contributing to memory for space, a complex form of explicit memory storage.
The general finding that long-term plasticity and long-term memory recruit transcription in the nucleus, an organelle shared by all synapses of a neuron, has raised a question we have begun to explore in Aplysia and mice: Are long-term changes cell-wide, or can induced gene products be spatially compartmentalized, so that they selectively alter the function of some synapses and not others? In mice we have also explored the molecular mechanisms whereby attention modifies and stabilizes internal representation of space.

Kandel, M.D. By probing the synaptic connections between nerve cells in the humble sea slug, Eric Kandel has uncovered some of the basic molecular mechanisms underlying learning and memory in animals ranging from snails to flies to mice and even in humans. His groundbreaking studies have demonstrated the fundamental ways that nerve cells alter their response to chemical signals to produce coordinated changes in behavior. This work is central to understanding not only normal memory but also dementia and other mental illnesses that affect memory. Kandel's research has shown that learning produces changes in behavior by modifying the strength of connections between nerve cells, rather than by altering the brain's basic circuitry. He went on to determine the biochemical changes that accompany memory formation, showing that short-term memory involves a functional modulation of the synapses while long-term memory requires the activation of genes and the synthesis of proteins to grow new synaptic connections. For this work, the Austrian-born Kandel was awarded the 2000 Nobel Prize in Physiology or Medicine. The traumatic events of Kandel's childhood likely influenced his later interest in the biological mechanisms of memory. He was only eight when, in 1938, Nazi Germany annexed his homeland, but the humiliation and discrimination that Kandel, his family, and other Jews suffered under this oppressive regime were forever seared into his memory. In 1939, on the eve of World War II, his family fled Austria for the United States.

In invertebrates the focus of our research is on the gill-withdrawal reflex of Aplysia. We study three elementary forms of learning: habituation, sensitization, and classical conditioning. Recently we have reconstituted critical components of this learning in dissociated cell culture, and we now use the reconstituted system to examine the molecular mechanisms which contribute to short- and long-term memory. In vertebrates we use genetically modified mice to examine the mechanisms of long-term potentiation in the mammalian hippocampus and its relation to spacial memory and maintenance.

Eric R. Kandel, M.D. (Distinguished Investigator 2000) of Columbia University, is undertaking a new series of studies on the molecular mechanisms in the amygdala that govern the learned acquisition of anxiety, and the delineation of genes recruited during the acquisition of fear. Context conditioning of fear will be examined as a model of chronic anxiety and cued conditioning of fear as a model of anticipatory anxiety. The function of genes expressed during these two types of anxiety states will be determined using methods for regional gene expression restricted to the amygdala, behavioral analysis, and analysis of a phenomenon known as long-term potentiation in the amygdala. This work has promise for elucidating the neural circuits and genes that cause anxiety disorders.
