Se graduó en la Universidad de Utrecht e ingresó como profesor de fisiología en la de Leiden, donde permaneció desde 1886 hasta su muerte. En 1903 desarrolló el galvanómetro que lleva su nombre, gracias al cual logró medir las diferencias de potencial eléctrico experimentadas por el corazón durante las contracciones sistólicas y diastólicas y reproducirlas gráficamente. Este procedimiento, que llamó electrocardiograma, le permitió descubrir que diferentes afecciones cardíacas producían distintos registros. Entre 1908 y 1913 estudió el funcionamiento de corazones sanos para así delimitar un cuadro de referencia, gracias al cual estudiar las desviaciones provocadas por la enfermedad. Hopps was trained as an electrical engineer at the University of Manitoba and joined the National Research Council in 1941, where he conducted research on hypothermia. While experimenting with radio frequency heating to restore body temperature, Hopps made an unexpected discovery: if a heart stopped beating due to cooling, it could be started again by artificial stimulation using mechanical or electric means. Ez a muszer, mint gyakorlati klinikai berendezés, a szívbetegségek diagnosztizálásának fontos eszköze lett. Az Utrechti Egyetemen szerzett orvosi diplomát, majd a Leideni Egyetem élettanprofesszoraként dolgozott 1886-tól egészen haláláig. 1903-ban tervezte meg az elsô húros galvanométert - ma Einthoven-féle galvanométernek nevezik -, ezzel a szívizom összehúzódásakor keletkezô elektromos potenciálváltozásokat lehetett mérni és grafikus úton rögzíteni. Einthoven felismerte, hogy a különféle szívbetegségekrôl nyert felvételek görbéi között eltérések vannak. 1908 és 1913 között az egészséges szív tevékenységérôl készült felvételek hullámait tanulmányozta avégett, hogy a kóros jelenségek pontosan felismerhetôek és értelmezhetôek legyenek. Einthoven továbbfejlesztette az elektródák elhelyezési rendjét, és a manapság alkalmazott standard (végtagi) elvezetések leírása és használata eredetileg az ô nevéhez fuzôdik.
