John D. Rockefeller, Jr. Professor; Investigator, HHMI The unidirectional translocation of thousands of distinct proteins across specific intracellular membranes is mediated by "signal" sequences. On average, a signal sequence consists of a stretch of ~15 amino acid residues that is either a transient or permanent part of the protein to be translocated. The signal sequence functions essentially as a ligand. Each signal sequence is membrane specific and is decoded by a complex machinery that is restricted in its location to one particular cellular membrane. The mechanism of protein translocation across the nuclear pore complex (NPC) is distinct from that of translocation across the above-described protein conducting channels. NPCs are huge organelles (estimated molecular mass: 125 million daltons) that are suspended in 100-nm wide circular openings in the nuclear envelope. An NPC can open to 25 nm in diameter. For passage across, proteins do not need to be kept in an unfolded configuration. Unlike protein conducting channels, NPCs are unable to integrate proteins into the lipid bilayer. Furthermore, transport across the NPC is bidirectional. Also, transport is not limited to proteins but includes ribonucleoproteins (RNPs). An in vitro system for signal sequence-mediated protein uptake into the nucleus has been used to isolate and to characterize cytosolic factors that are required for import. Similar

1945 was also a turning point in my life. Until then my childhood was a perfect 19th century idyll. In the cold and snow-rich Silesian winters there were hour-long rides on Sundays in horse-drawn slays to my maternal grandparent's farm to have lunch and to spend the afternoon. The house was a magnificent 18th century manor house in the nearby Altgabel with a great hall that was decorated with hunting trophies. In the summer, of course, horse-drawn landauers were used as means of transportation. The way to school was a long one. We went there on foot and as a pack, usually consisting of one or two of my seven brothers and sisters and of children from neighboring houses.
The months before and after the end of World War II were chaotic and miserable. None of my relatives had enough space to accommodate our large family leaving us divided among several relatives in different villages. There was no communication and little food. On September 9, 1945, we learned of the death of my beautiful oldest sister Ruth who, at age 19, was killed in an air raid on a train she was travelling in on April 10, 1945. She was buried in a mass grave near the site of the attack in Schwandorf, Bavaria. Ruth was born when my mother was just 20. The two had a sisterly relationship. My mother grieved over Ruth's death until the end of her own life.

Midwood High School at Brooklyn College, Brooklyn Cells are bound by lipid bilayer membranes that have many proteins between them. Membranes composed of proteins and lipids enclose many organelles that perform important life processes in the cell. The endoplasmic reticulum (ER) and the ribosomes synthesize proteins. Proteins play various important roles within cells, such as forming the composition of the cell, and catalyzing chemical reactions as enzymes. Such findings helped to enhance the knowledge of cellular processes (Gallwitz). Under the mentorship of George E. Palade (Nobel Prize Laureate in 1974), who had already found the intracellular path of secretory proteins, Blobel began to perform intense research at Rockefeller University in New York into the issue of cellular protein targeting. Palade had already found that secretory proteins traverse the rough endoplasmic reticulum (ER) membrane, where they are modified after which they enter the Golgi Apparatus for packaging, before being exported from the cell. The biochemical/molecular mechanisms were unknown at that time, and Blobel attempted to explain this process by these means. For his great discoveries, Blobel was awarded the Nobel Prize in 1999 (Gallwitz; Press Release: The 1999 Nobel Prize in Physiology or Medicine; Blobel 86-102).
