Im Jahre 1942 wurde er als Sohn italienischer Einwanderer in New York geboren. Bereits als Kind litt er aber an Asthma und musste die meiste Zeit zu Hause verbringen - ein Grund für ihn, am liebsten vor dem Fernseher zu hängen. Auch im lokalen Kino wurde er schnell Stammgast, es gab kaum einen Film, den er nicht sah, die angeschlagene Gesundheit machte ihn geradezu süchtig nach Filmen. Mit 14 Jahren beschloß er allerdings, Priester zu werden. Zum Glück für die Cineasten dieser Welt wurde er im College seiner Wahl nicht aufgenommen - so wurde für ihn der Weg frei, um zu seiner tatsächlichen Berufung zu finden: dem Filme machen. Der Italo-Amerikaner Martin Scorsese absolvierte sein Studium an der New York University und schrieb sich währenddessen für die Filmkurse ein. Er arbeitete nach dem Studium als Cutter und drehte einige Kurzfilme, die bereits Auszeichnungen erhielten. Sein erster Langfilm Who's That Knocking At My Door (1969) mit Harvey Keitel erregte die Aufmerksamkeit von Roger Corman , der ihn bat, bei Die Faust der Regel ("Boxcar Bertha", 1972) die Regie zu führen.
